"In recent years we have learned that many cases of depression are actually caused by an imbalance of substances called neurotransmitters, chemicals in the brain that are intricately involved in how we think and feel. For example, Prozac, probably the best known of the antidepressants, works by restoring the normal balance of a neurotransmitter known as serotonin. In addition to performing the routine functions of regular hormones, we now know that superhormones are also similar to neurotransmitters; they are located in the brain and have a profound impact on mental function. That is why we believe that many cases of depression in older people are actual caused by the decline in superhormones and, in particular, pregnenolone and/or DHEA. I believe that restoring the superhormones such as pregnenolone to their youthful levels will prevent many of the cases of depression that occur among older people and that are all too frequently written off as part of the "normal" aging process.
My belief has been confirmed by a recent study conducted by the National Institutes of Mental Health; it showed that people with clinical depression have lower than normal levels of pregnenolone in their cerebral spinal fluid (the fluid that bathes the brain). Although this study did not look specifically at older people, this finding is relevant to anyone who is producing lower than normal levels of pregnenolone, and that is precisely what happens in aging. What is the connection between pregnenolone and depression? Let me explain the theory. The nerve cells in our brains are constantly being bombarded with stimuli and receiving and transmitting information. Our brains contain a neurotransmitter called GABA, which literally cools the brain and protects our nerve cells from burning out in the course of all this activity. GABA performs an important function, but too much GABA may make the brain too sluggish and produce a dampening effect that depresses brain function. It appears that pregnenolone mitigates the effect of GABA and restores balance to the brain. You may remember that in the earlier studies performed on pregnenolone in the 1940s, the researchers noted that the participants reported an enhanced sense of well-being and an improvement in mood. This could also be due to pregnenolone's effect on GABA.
Although there have been no official studies done on the treatment of depression with pregnenolone, based on all we know about this superhormone, I believe that in cases of mild depression pregnenolone should be very helpful and certainly should be tried before bringing on the "heavier guns," stronger medication that can cause substantial side effects. We know that pregnenolone is safe, well tolerated, and causes no known side effects, so it just makes good sense to try it first.

An Historical Treatment for Arthritis?              
In the 1940s, pregnenolone was used -----as a treatment for rheumatoid arthritis, the disease that causes severe joint pain and fatigue, and that continues to plague millions of sufferers. As you may remember from the chapter on DHEA, rheumatoid arthritis is an autoimmune disease that is more likely to strike as we get older. There is as yet no cure for rheumatoid arthritis, and the symptoms are usually treated with anti-inflammatory medication. Nearly a half-century ago, rheumatoid arthritis patients  took pregnenolone  and reported that they felt less pain and were less tired, and much stronger.  Was it just the placebo effect? Despite the fact that pregnenolone showed promise as a treatment for rheumatoid arthritis, all research on pregnenolone as an arthritis treatment ended abruptly in the 1950s. One reason the medical community lost interest was that, like the other superhormones, pregnenolone was a natural substance produced by the body and therefore could not be patented. As a result, no drug company was interested  in investing the time or money to bring pregnenolone to market. As previously mentioned, it is a sad fact of life that in the United States, no matter how effective a substance may be in treating illness or extending and improving the quality of life, if it is not patentable, it stands a good chance of falling by the wayside. Again, that was a primary reason for writing The Melatonin Miracle and for writing this book. I hope to awaken public interest in these extraordinary natural substances; they are doomed to oblivion unless someone steps forward (I feel it must be the government) to fund these important studies.

There was another reason that pregnenolone was forgotten: It was completely overshadowed by the discovery of an exciting new synthetic cortisone, in the late 1940s; it was thought to be the cure for arthritis. When cortisone was first introduced, it received as much attention  as did penicillin when it launched the antibiotic revolution a decade earlier. Unlike pregnenolone, the effects of which were cumulative and felt over a period of weeks, cortisone also produced almost overnight relief from arthritis symptoms. From the point of view of pharmaceutical companies, which could make real money selling cortisone, and of patients who understandably wanted fast, fast, fast relief, cortisone was a dream come true. The problem is that by the time we woke from the dream and realized that cortisone and its related compounds have some pretty nightmarish side effects-such as a dampening effect on the immune system that makes patients susceptible to infection, and a destructive effect on bone mass, leaving patients vulnerable to osteoporosis-we had forgotten that once upon a time there was a substance called pregnenolone, which produced relief from ------ symptoms much more safely, albeit more gradually.
